Parkinson‟s disease (PD) is a neurodegenerative disorder of the mid brain region of the central nervous system for which no neurorestorative therapeutic treatment is currently available. Oxidative stress along with neuroinflammation plays an significant role in the pathophysiology of PD. The earliest Indian medical system, Ayurveda, conventionally uses Mucuna pruriens (Mp) and Withania somnifera (Ws) to treat PD. Recently, Mp and Ws has been shown to possess antiparkinson and neuroprotective effects in various animal models of PD. The antioxidant activity of Mp and Ws was suggested by its capability to scavenge various free radicals and reactive oxygen species. Mp and Ws significantly inhibited the oxidation of lipids and deoxyribose sugar. Mp and Ws also exhibited metal chelating activity and did not exhibit any genotoxic/mutagenic effect on the plasmid DNA. These interesting results propose that the neuroprotective and neurorestorative effect of Mp and Ws may be connected to its antioxidant and anti-inflammatory activity independent of the symptomatic effect. In addition, Mp and Ws also exhibit synergistic effect in PD mouse model. Thus in this review article, magical role of Mp and Ws has been discussed.
