Abstract
HEPATOPROTECTIVE EFFECT OF METHANOL LEAF EXTRACT OF SMILAX KRAUSSIANA ON CARBON TETRACHLORIDE – INDUCED TOXICITY IN RATS.

ABSTRACT

Objectives: The decoction of leaf extract of Smilax krausiana in hot water or alcohol is employed locally by Uruan indigenes of Akwa Ibom State, Nigeria, in the management of poisoning. The aim of this investigation is to ascertain whether it can ameliorate carbon tetrachloride – induced toxicity in rats, and through which mechanism it mediates its effect. Methods: The leaf was harvested from the wild, identified and authenticated by Taxonomist. It is later macerated for 72 h in methanol, filtered and dried to a constant weight. The rats were divided into seven groups containing six rats per group. Groups 1 & 2 were treated with normal saline (10 ml/kg, p.o). Group 3 received silymarin (100 mg/kg, p.o). Groups 4-6 were treated with 72, 144 and 216 mg/kg respectively while group 7 was treated with extract 144 mg/kg (p.o), 10min later, silymarin was administered. All the groups were administered with their respective dosages for seven days. On 8th day, the animals in groups 2-7 were administered with carbon tetrachloride (0.7 ml.i.p) Twenty four (24h) hours later, the animals in all groups were weighed and sacrificed under light ether anaesthesia and the samples collected for analysis. The antioxidant effect of the extract was also investigated. Result: The extract dose-dependently reduced the CCl4 - induced toxicities in ALT, ALP and cholesterol. Others were Pr, Hb and PCV. It also reduced in a non dose – dependent manner the effects of CCl4 on AST and LDH. These reductions were statistically significant (p<0.001). The activity of MDA decreased as the concentration of the extract increased. Converse was true for DPPH. The effects of the CCl4 on organs of localization were maximally reduced. Conclusion: The ability of the extract to exhibit hepatoprotective effect against CCl4 – induced toxicity is inherent in the phytochemical properties (polyphenols) of the plant. These properties therefore lent credence to its folkloric use in the treatment of poisoning.
