A STUDY BASED ON AUGMENTATION IN THE ACTIVITY OF TACROLIMUS PRODUCING MICROORGANISM

ABSTRACT

Tacrolimus is an immunosuppressive drug designed to foster medical assistance in case of organ transplants and treatment of autoimmune disorders. Very few organisms have been studied in regard of tacrolimus production. This secondary metabolite is produced as part of a multifaceted biosynthetic pathway of the organism. For the current study, the actinobacteria named Streptomyces tsukubaensis has been considered to obtain tacrolimus in substantial amounts. The microbe has been cultured in YMA medium slants to aid in single colony isolation. Combinatorial as well as individual studies encompassing the effect of various medium components has been an integral part of the present investigation. Plackett-Burman Design and One Variable At a Time (OVAT) approaches have been applied to optimise the production media. The consecutive methodology adopted has been found to be proficient in enhancing the activity from 200 mg/L to 500 mg/L while citing interesting insights on the significant variables, namely dextrin white and polyethylene glycol (PEG).
