FORMULATION DESIGN, EVALUATION, AND OPTIMIZATION OF CIPROFLOXACIN HCL FLOATING TABLET USING DESIGN EXPERT

ABSTRACT
Ciprofloxacin HCl is a widely prescribed antibiotic, but its oral bioavailability can be compromised due to its limited solubility and absorption in the stomach. Floating tablets offer a promising solution to this problem by prolonging the drug's residence time in the gastric region. This study aimed to design, evaluate, and optimize a floating tablet formulation of Ciprofloxacin HCl using the Design Expert software. full factorial design was employed, with two independent variables, namely, the amount of sodium bicarbonate and the concentration of HPMC K4M. The dependent variables included the tablet's floating lag time, total floating time, drug release at 12 hours, and drug release at 6 hours. The results indicated that sodium bicarbonate and HPMC K4M had significant effects on the tablet's floating behavior and drug release. By using the Design Expert software, an optimized formulation was generated, and the desirability function was maximized. The optimized formulation exhibited a short floating lag time, prolonged floating duration, and controlled drug release, which can enhance the drug's bioavailability and therapeutic efficacy. In conclusion, this study demonstrated the successful formulation design, evaluation, and optimization of Ciprofloxacin HCl floating tablets using Design Expert. The optimized formulation showed promising attributes for controlled drug delivery, suggesting its potential application in the treatment of gastric infections. Further in vitro and in vivo studies are needed to confirm the clinical effectiveness of the developed formulation.
