FORMULATION, DEVELOPMENT AND EVALUATION OF COLON TARGETED DELIVERY OF INDOMETHACIN

ABSTRACT
Colorectal cancer is a significant cause of cancer-related deaths in developed nations. Indomethacin, a promising anticancer NSAID, faces challenges due to severe GI side effects with oral administration. This study explores novel formulations aiming to deliver indomethacin to the colon while minimizing GI exposure. Traditional pH-sensitive polymer coatings have inconsistent performance. This study proposes matrix-based systems combining pH and transit time control. Preformulation studies guided polymer selection. Various single and multi-unit systems were developed, demonstrating controlled release patterns in vitro. In vivo rat studies confirmed pH and transit time-dependent release, with promising outcomes for some formulations, necessitating further optimization. In conclusion, matrix-based systems offer a potential solution for targeted indomethacin delivery to the colon, addressing GI toxicity concerns in colorectal cancer treatment.
