ANTIBIOFILM EFFECT OF ASCORBIC ACID AGAINST DUAL BACTERIAL BIOFILMS OF SALMONELLA SPECIES WITH DIFFERENT BACTERIA

ABSTRACT
Numerous food-borne pathogens such as Salmonella, Staphylococcus, Pseudomonas and E. coli were proven to co-exist in food processing facilities resulting in formation of multispecies biofilms leading to food spoilage and/or food poisoning. Thus, there is urgent need for effective antibiofilm agent targeting poly-microbial biofilms. Hence, in the present in vitro study, Ascorbic Acid was evaluated for its Antibacterial and Antibiofilm potential against total 9 dual bacterial species of S. enterica and S. typhi with different Bacteria using MIC, MBC and Crystal violet staining method. Additionally, combination prepared from Ascorbic Acid (AA) and Ciprofloxacin (CP) standard antibiotic was evaluated to determine its combinatorial effect against total 9 Dual bacterial biofilms. Ascorbic Acid exhibited the highest antibiofilm activity at the concentration of 10 mg/ml against the dual bacterial biofilms in the following order, S. typhi + MRSA (84.33%) > S. typhi + P. aeruginosa > S. typhi + E. faecalis > S. enterica + E. faecalis > S. enterica + MRSA > S. enterica + P. aeruginosa > S. enterica + S. typhi (38.32%). However, Ascorbic Acid did not show antibiofilm activity against dual bacterial biofilms of S. enterica + E. coli and S. typhi + E. coli at the same concentration. Interestingly, the combination AA+CP displayed the highest antibiofilm activity against dual bacterial biofilm of S. typhi + E. coli (88.19%). In our previous studies, Ascorbic Acid has displayed antibiofilm potential against Mono and Mixed-Bacterial Species as well as against C. albicans biofilms and multispecies biofilms of C. albicans with Mixed Bacteria (Fungus + Bacteria), whereas, in present in vitro study, Ascorbic Acid has revealed antibiofilm potential against Dual bacterial species (Bacteria + Bacteria).Therefore, Ascorbic Acid could be a promising candidate with Antibiofilm potential to treat polymicrobial biofilms in food industry or hospital-acquired infections.
