FORMULATION AND EVALUATION OF BUCCAL ADHESIVE TABLET OF PROMETHAZINE HYDROCHLORIDE USING NATURAL POLYMERS

ABSTRACT
The present study aimed to formulate and evaluate buccal adhesive tablets of Promethazine Hydrochloride using natural polymers such as HPMC, Carbopol, and sodium alginate to achieve sustained drug release and improved bioavailability. Preformulation studies confirmed the purity and suitability of the drug for formulation. Tablets were prepared by direct compression and evaluated for pre- and post-compression parameters, swelling index, and in vitro drug release. All formulations showed satisfactory physicochemical properties and mucoadhesive behavior. Among them, formulation F5 exhibited optimal swelling characteristics and sustained drug release (98.15% over 12 hours), following first-order kinetics with a non-Fickian release mechanism. The optimized formulation demonstrates potential as an effective buccal delivery system for prolonged therapeutic action and enhanced patient compliance.
