AMELIORATION EFFECT ON DIABETES BY ETHYL ACETATE AND METHANOL LEAF FRACTIONS OF Nauclea Latifolia IN ALLOXAN-INDUCED DIABETIC RATS

ABSTRACT
The antidiabetic effects of ethyl acetate and methanol fractions of Nauclea latifolia were investigated in alloxan induced diabetic rats. Fractions of Nauclea latifolia at the doses of 150 and 300mg/kg were administered orally to the experimental animals for fourteen days while their blood glucose levels were taken daily. At the end of the experiment their lipid profile were assayed. The analysis of the results showed that there were significant decreases (P=0.05) in the blood glucose in both fractions (150mg/kg of ethylacetate-150.50+31.44mg/dl; 300mg/kg ethylacetate-134.25+28.62mg/dl; 150mg/kg methanol-163.75+93.65mg/dl) except for the 300mg/kg methanol fraction (368.67+79.82mg/dl) when compared to the untreated diabetic group (394.66+84.47mg/dl). The lipid profile assay present a significant reduction (P=0.05) in serum TC (150mg/kg of ethylacetate-69.88+8.52; 300mg/kg ethylacetate-51.08+9.93mg/dl; 150mg/kg methanol-116.86+13.34mg/dl; 300mg/kg methanol 108.66+12.77mg/dl) and LDL-C (150mg/kg of ethylacetate-8.23+7.76mg/dl; 300mg/kg ethylacetate-6.44+7.66mg/dl; 150mg/kg methanol-50.68+14.13mg/dl; 300mg/kg methanol-42.09+79.93mg/dl) when compared to the untreated diabetic group (TC=383.76+79.68mg/dl and LDL-C=299.46+79.23mg/dl). Also, the 150mg/kg and 300 mg/kg ethylacetate groups significantly (P=0.05) reduced the LDL-C when compared with the group treated with the standard drug (32.06+13.23mg/dl). Similar reduction was observed in VLDL-C of fractions (150mg/kg of ethylacetate-34.94+1.84 mg/dl; 300mg/kg ethylacetate-22.24+2.77mg/dl; 150mg/kg methanol-35.13+2.78mg/dl; 300mg/kg methanol-33.76+3.17mg/dl) in comparison to the untreated diabetic group (42.58+10.35mg/dl). Thus, fractions of Nauclea latifolia ameliorates diabetes mellitus: ethyl acetate fraction may be a better agent in arresting hyperglycaemia and hyperlipidemia arising from diabetes mellitus when compared to the methanol fraction; in a dose dependent manner where the higher dose is more significantly efficacious.
