GELUCIRE IN DRUG DELIVERY SYSTEMS

ABSTRACT
Lipids based drug delivery system is a multidisciplinary approach, a assembly of organic chemistry, physics, biopharmaceutical and formulation technologies encompassing the role of fatty acids, fatty acid esters, and their combinations thereof, in order to improve release and drug absorption. Gelucires are a novel class of synthetic polymers or vehicles obtained from mixtures of mono, di- and triglycerides with polyethylene glycol (PEG) esters of fatty acids. Gelucires containing only glycerides or a mixture of glycerides and PEG esters (Gelucire 54/02, 43/01) are used in preparation of sustained release formulations, while Gelucires containing only PEG esters (Gelucire 55/18) are generally used in the preparation of fast-release formulations. The objective of this study was to explore the potential applications of gelucire in controlled drug delivery systems. Gelucire are available for commercial use according to their HLB values and different melting points. Various approaches to receive controlled drug delivery systems like gastro retentive floating drug delivery system, sustained release dosage forms, improving the pulmonary absorption of drug, & to achieve SMEDDS using gelucire are explained. Here we aim to evaluate the use of gelucire as a non-ionic surfactant to enhance the bioavailability of various drug models. From this study it is concluded that use of gelucire can be considered as an effective carrier and indomitable excipient for the design of drug delivery system for a wide variety of ingredients.
