ABSTRACT

Osteoporosis is an important clinical problem following menopause and old age. Most of the bone loss is attributed to genetic factors associated with osteoclastic activity. The most common prescription for osteoporosis is the supplementation of calcium and vitamin D. Examining the basic mechanism for bone formation and mineralization, shows that supplementation with basic bone mineralizing nutrient; L-Arginine, L-Proline, L-Lysine, L-Ascorbic acid, Zinc would be helpful in taking care of collagen synthesis and bone formation and would be a better option to sole calcium/vitamin D supplementation (Treatment 1, T1). Vitamin K2 has also been reported to help in the bone formation and mineralization process. We thus formulated T2 by supplementing vitamin K2 to T1. The trigger for bone loss in women is the deficiency of estrogen. We supplemented phytoestrogen, genestein isoflavone to T2 to formulate T3. Treatments T1, T2 and T3 were orally given to ovariectomised female wistar rats for 100 days. At the end of the treatment period, rats were sacrificed by CO2 asphyxiation. The biochemical parameters including serum calcium, phosphorus, lipid profile, urine calcium, createnine and phosphorous, shows improved status in treatment group as compared to control, histological studies shows increase in thickness of bone trabecular network, and improved bone mineral content determined by ICP-AES. Our results encourage dietary intervention with bone mineralizing nutrients could be a supplementary therapy for postmenopausal osteoporosis.

Keywords: osteoporosis, Basic bone mineralizing nutrients, bone health, collagen, ovariectomy.
