DESIGN AND DEVELOPMENT OF CHITOSAN NANOPARTICLE IN TARGET NOVEL DRUG DELIVERY SYSTEMS FOR UROLITHIASIS

ABSTRACT
Nanoparticles based drug delivery has been recognized to improve the solubility of poorly hydrophilic drugs, to prolong the half-life of drug systematic circulation and releases drugs at a sustain rate. This present research work deals with the inhibitory potential of Aerva lanata water extract and plant extract loading Chitosan nanoparticle were tested for crystal formation in kidney under in vitro conditions. In vitro Antiurolithiatic activity comprises nucleation, growth and aggregation of calcium oxalate crystals were measured separately by UV- vis Spectrophotometric methods. The nanoparticle loaded with Aerva
lanata leaf extract were synthesized using ionic gelation method. The nanoparticle formulation shows poly-shaped with small in size and it has high loading efficiency. Plant extract loaded nanoparticle were prepared, and their maximal encapsulation efficiency was up to (98.11±4.9%). In vitro release showed that this nanoparticle provided an initial burst release followed by a slowly sustained release for more than 24 h. The drug releasing chitosan nanoparticle plays a major role in Aerva lanata extract delivery to the kidney and is achieved at a near neutral (pH=7.4) condition. The synthesized nanoparticles were characterized using Scanning Electron Microscope (SEM) and Fourier Transform Infrared Spectroscopy (FTIR) analysis. These nanoparticle were 20-500 nm in size and carried obvious positive surface charges.
