Abstract
APPLICATION OF RANDOM AMPLIFIED POLYMORPHIC DNA MARKERS FOR AUTHENTIFICATION OF THREE DIFFERENT AVAILABLE SOURCE PLANT OF A FREQUENTLY USED AYURVEDIC DRUG - TRIVRUT.

ABSTRACT

Trivrut, known as Nishotha in Indian herbal market, is one of the important and frequently used herbal drug in Ayurveda. It has been considered as best among the laxative drug (Sukhavirechana). Differences in the opinion regarding types of Trivrut, its synonyms have made Trivrut as controversial drug. Operculina turpethum Silva
Manso (Convolvulaceae) and Marsdenia tenacissima Wight & Arn.(Asclepiadaceae), are being used as a Trivrut. Root of Operculina petaloidea Choisy (Convolvulaceae) is also considered as Shyama Trivrut. Pharmacognostical study of individual as well as comparative study of three source drugs has been published, but till date, no work has been reported on their DNA analysis. In the present study, molecular characterization of the plant by Random Amplified Polymorphic DNA (RAPD) markers was studied following standard parameters. Genomic isolation of DNA from fresh leaves was amplified by RAPD markers. The unique bands obtained in Polymerase Chain Reaction (PCR) amplification were clearly discriminated having, many bright and light bands indicating the genuinity of the plant. Results were subjected to Jaccard coefficient band scoring and analysis with clustering, 60% similarity between sample O turpethum and O petaloidea and 40% similarity was observed in sample O turpethum and M tenacissima.
